Targeted Therapies

•   Nomenclature:

-   Small molecule inhibitors end in “-inib”

-   Monoclonal antibodies end in “-mab”

•   Tyrosine kinase inhibitors: Small molecule inhibitors, given orally

-   Imatinib (Gleevec), dasatinib: Bind to the inactive form of BCR-ABL and prevent activation. Used for treatment of CML.

-   Osimertinib, erlotinib: EGFR inhibitors used in NSCLC

-   Sorafenib, sunitinib: VEGF inhibitors used in HCC, RCC, thyroid

•   Monoclonal antibodies: Given via infusion

-   Anti-HER2: Trastuzumab (Herceptin), pertuzumab (Perjeta) used in breast cancer. Trastuzumab also used in esophageal/gastric cancer.

-   Anti-CD20: Rituximab, targets B-cells, used in B-cell lymphomas

-   Anti-VEGF: Bevacizumab (Avastin), inhibits angiogenesis, used in colon cancer, RCC
